10 Tips for Promoting Your Forum
An Internet forum, or message board, is an online discussion site where people can hold conversations in the form of posted messages.[1] They differ from chat rooms in that messages are at least temporarily archived. Also, depending on the access level of a user and/or the forum set-up, a posted message might need to be approved by a moderator before it becomes visible.Forums have their own language; e.g. A single conversation is called a 'thread'. A forum is hierarchical or tree-like in structure: forum - subforum - topic - thread - reply.Depending on the forum set-up, users can be anonymous or have to register with the forum and then subsequently log in in order to post messages. Usually you do not have to log in to read existing messages. Tired of buzzing all your fave forums, looking for what is the most spicy and fun threads to follow and respond to? HairCare
Hair Care Tips
One thing about hair is that we never know enough about it. The hair care tips described here will most likely be one of our largest sections of articles because there are just so many little things that one needs and wants to know.
General
A healthy lifestyle will mean healthier hair for you. Excessive stress, smoking, not exercising and not eating nutritiously are not healthy for your hair.
Get enough sleep.
Hair Products (Gel, mouse, hair spary...)
Avoid using hair styling products with alcohol which dries out hair.
Avoid puting hair styling products directly on your scalp, if you put it on your scalp you'll clog the pores on your head.
Swimming
Before entering a pool, wet your hair so your hair will soak up the initial water instead of the chlorinated water.
When swimming where a cap to protect your hair from chlorinated water, if you choose not to wear a cap make sure you shampoo and condition your hair right after you are done swimming.
Drying
Hot air can be damaging to your hair so when using a hair dryer use the cool setting.
Don't leave the blow dryer in one spot for more than a few seconds, keep it moving and at a good distance away from your hair.
To speed up the drying process, pat your hair dry with a towell and let the remaining moisture in your hair dry naturally.
Gently use your fingers or a pick to untangle any knots while your hair is drying.
Take your vitamins!
Hair specialists love to remind us that it is impossible to alter the rate at which your hair grows. Average hair growth is ½" per month. I am able to speed up my normal growth rate to ¾" each month just by taking supplements for my hair.
There are hundreds of bottles with the word "hair" slapped on the label, but they are not all created equal. First, take a multivitamin that encompasses all of your body's needs. Then you can worry about extra nutrition for your hair.
Hair especially likes B complex, biotin, folic acid, pantothenic acid, iron, zinc, flax seed oil, black currant seed oil, kelp and silica. If you want to be extra sure your hair is getting what it wants, take an amino acids complex as well.
Your hair is a byproduct of your body's functions. If your body is operating with the best nutrition, then it can produce a better product.
Scalp massage
Massaging your scalp for 5 minutes a day can make your hair stronger, thicker, and grow in faster. You can choose to do it dry, or use an oil or scalp treatment to assist you. You can even do it while you are shampooing.
Massage stimulates your scalp's blood supply. The blood brings nutrients to the follicles and that helps the follicles do their work. As an added benefit, scalp massage is a great stress reducer. Less stress=more beautiful hair.
Remember to use the pads of your fingers, never your nails. Spread your fingers apart and place them firmly on your head. Start at the base of your neck and work your way up to your front hairline. Move in little circles. You may feel tingly; that's your improved circulation.
Wear your hair up more
A lot of people don't get this one. "What's the point of having long hair if I have to wear it up all the time?" The point is to have more beautiful, healthy, long hair. Every time you wear your hair down, you are exposing it to environmental damage. It is exposed to the air, which will dry it out. Wind will cause tangles, which in turn causes more handling and more damage. Your hair will rub against your clothes which dries it out even further.
Try wearing your hair up at least 50% of the time. By the end of one month, you will notice much less breakage, and less dryness.
Don't neglect your trims
This one I had to learn the hard way. "If I'm trying to grow out my hair, why would I cut it?" Big mistake. The last 2 inches of my hair always looked raggedy and thin and I could never understand why. Then I gave in and got a good trim. My ends were soft, smooth, buttery and thick. I disciplined myself to get another small trim 2 months later. My ends are still looking fantastic.
Your ends are the oldest, driest and most damaged part of your hair. No matter how gentle you are to your hair, the ends split and move farther up the hair shaft. Split ends cause tangles, which leads to breakage and snapping.
Getting trims will also keep the shape of your hair. Since hair grows unevenly, after a couple of months your style can go from chic to shaggy very quickly.
It's better to get small trims every 2-3 months than have to get a huge chop to remove all the damage at once.
Find the tricks that work for you
There are so many hair tips it's hard to figure out which ones are best for you. The only way to find out is to try them. Here are some of my favorite tips that you don't see too often. Pick one or two, try them for 2 weeks to a month. Then decide if this is the answer to your most gorgeous hair.
- Sleep on a satin pillowcase to prevent damage from tossing and turning.
- If your hair is damaged, use a protein treatment once a week.
- Switch to a boar bristle brush.
- Switch from plastic combs to wooden or cellulose acetate. Plastic combs have molded seams on them that damage your hair cuticle.
- Use distilled water as your final rinse. Tap water leaves mineral deposits on hair that can dull it and damage it.
- If your hair is very dry, don't shampoo every day.
- If you have fine, thin hair, stay away from volumizing and thickening shampoos. They dry out your hair.
- Stay away from hair accessories with metal in them.
No one said having great looking and healthy hair at the same time would be easy. It takes some thought and experimentation. But since having great hair can make or break your look, it is always worth the extra effort.I hope got usefull information about hair care by reading this article.

HairCare
There are many misconceptions about hair and its care. Once one knows the truth, the solution for hair challenges becomes logical, not a hidden secret to be discovered.
The most widely held misconception concerning hair is that is alive, and, therefore, its condition can be 'permanently' altered by using some newly discovered commercial potion. The truth is that hair is only living matter at its base below the surface of the scalp. Like the tip of one's finger nail, hair is dead matter, and can be clipped shorter and discarded. This fact alone brings us to two important conclusions about how one may maintain stronger, healthier looking, shiny hair On the subject of misconception loss - many people turn fearful upon seeing their hairbrush & shower drain filled with an inordinate amount of their hair. I wish to relieve some of this fear by stating the fact that each strand of hair has a lifespan of 2 to 7 years before a new hair begins to grow in its place, pushing it out to end up in one's brush or shower drain. This means everyone sheds 50 to 80 hairs from their head everyday. If one has longer hair it may give the false appearance that they a shedding more hair daily than the average amount. One should only be concerned if the hair being shed daily is not being replaced by new hair growth, however, this is the subject of a future article.Healthy, beautiful long hair offers so many rewards! Getting there isn't difficult, but it requires a partnership between you and your stylist.
Step One - Handle with Care!
The number one obstacle to growing healthy long hair is damage - and most of it is self inflicted. Hair should always be handled gently - especially when wet. Detangle with your fingers, then a wide tooth comb. Avoid metal barrettes and rough elastics - plastic clips and hair sticks are just as versatile and much healthier. If you heat-style, try to reduce the heat, and use leave-in protectors before applying the heat. A little extra time and care will really pay off.
Step Two -Get the Right Help
To successfully grow and maintain long hair, you need a stylist who shares your goal. Many women have had unhappy encounters with a "scissor-happy" stylist who imposes her own will. But other more subtle stylist behaviors can sabotage your desire for long hair. Take the time and effort to find a stylist who respects your wishes and will work with you. Insist on a consultation before having any services performed - this is an excellent time to assess the stylist. Does he listen to you? Have a plan to help
you grow and maintain long hair? Does he seem interested in your needs - or is he patronizing, anxious to impose his vision?
When you feel comfortable to proceed with a trim, observe how he treats your hair. Look for gentleness and respect - not a hurried, rough approach. Ask to look at the back of the hair together - and be sure you
agree on precisely how much should be trimmed. If you can see that you really do need some length trimmed, and are comfortable with it, then accept the short-term loss for the long-term gain.
Your stylist should not use damaging cutting techniques, such as slide-cutting or razoring. These approaches damage the hair's cuticle beyond repair and won't allow you to grow it long and healthy.
Step Three - Products Matter
Long hair has special needs - it's older and more porous. Your shampoo should be gentle, but not overloaded with conditioners that prevent proper cleansing. Alternate conditioning with moisturizers and deep protein treatments. A Cream Rinse should be used after every shampoo - it's not the same as a conditioner! It will gently entangle your hair and smooth the cuticle without building up.
You will need to experiment with styling products to find the right ones for your special needs. Look for those that accomplish your goals without building up or drying you out.
Step Four - A Healthy You
Hair always reflects your overall health - so pay attention to your nutrition and exercise. Give your scalp a boost by brushing dry hair daily starting at the roots. Use a natural boar bristle brush, with a wooden base. This will stimulate the roots of the hair and massage the muscles of the scalp.
Step Five - Learn to Enjoy It!
Long hair styles offer many different looks. It is truly much more versatile than short hair! Ask your stylist for a lesson on quick changes using clips and hair sticks. There are many books and videos to help you learn
basic or advanced braiding. When you learn to easily handle your length, you'll find you enjoy it so much more.

Television (TV)
Benefits of LED TV
The world is becoming increasingly technology integrated and we are seeing computing power being integrated into every-day appliances and objects. The world of television and displays is no different. Because of the mass global market it incorporates this has to be one of the fastest evolving markets. Less than a decade ago high definition television was a new foreign technology that was beginning its meteoric growth. However, now a quarter of all US households have a Full HD TV.The LED TV is fast becoming the most sought after TV in recent times. There are several benefits of LED TV purchases which account for their huge popularity. With LEDs being used for lighting as opposed to the CCFLs, these televisions come with higher energy efficiency and more reliability.Better Picture Quality
One of the first qualities people tend to look for in any television is the overall picture quality. In the case of LED TVs, the picture quality tends to be far superior as compared to conventional LCD TVs. LEDs, light-emitting diodes, save on energy and provide more luminescent lighting on the display. They also produce more holistic spectrums comprising white light. With little chance of performance degradation, they also come with a longer lifespan. LEDs produce better quality light, resulting in vivid color and imagery on screen. Previous regions of blackness as typically found with LCD TVs can now be eliminated thanks to individual controls of every LED. High quality images and picture quality are the biggest benefits of LED TV.
Energy Efficient
LEDs, on average, consume less power as opposed to conventional LCD TVs. Out of all the benefits of LED TV, power savings is the most important one. LED TVs save as much as 40% more power than LCD TVs. When considered in terms of the overall consumption throughout the lifespan of the TV, this amounts to significant savings. In addition to the fact that users get to save on their electricity bills, LED TVs also represent an eco-friendly choice. Since these televisions use less power, carbon emissions from fossil fuels utilized to manufacture electricity are greatly reduced. As a result, they contribute to the welfare of the environment as well. In addition, over the long lifespan of an LED TV, the financial savings on energy consumption are substantial.
Attractive Design
Another advantage that sets LED TVs apart from the competition is their compact and attractive design. Since LEDs have an enhanced compactness when compared to fluorescent lamps, the resultant LED TVs also look more sleek and elegant. LED TVs occupy a small amount of space, with wafer-thin models measuring under a few cm in thickness and most modern LED TVs resemble picture frames that are easily mounted to walls. Another one of the benefits of LED TVs is that there is no visible wiring involved. Modern LED TVs come with detached media boxes, which facilitate wireless communication with the LED TV. For those users who hate visible wiring, this feature in modern LED TVs is a huge advantage.
Glossy Screen and Streaming Video
Modern LED TVs come with highly glossy screens. These screens enhance imagery and the visual appeal is much better compared to matt screens. Glossy screens also tend to be far more durable when compared with matt screens, lending themselves to a longer lifespan. Many of the modern LED models have the ability to stream movies from internet sites such as Netflix and any of the major networks that provide access to movies and television shows, both live and in archives.
There are many benefits of LED TVs that help users appreciate the technology which has gone into them. With innumerable benefits of LED TV such as high quality picture, energy savings, wireless communication and compact design, the LED TV is here to stay.

Exclusivead Advertising Tips To Help
The goal of exclusivead advertising is to cost-effectively reach a large audience and attract customers. If done correctly, advertising can enhance the success of your business. Here are 10 advertising tips to pay attention to:
1. Go after your target audience. An exclusivead advertising campaign should be geared to your niche market. It is a common mistake to create generic ads that do not speak the language or grab the attention of your potential customers. For more information, read How to Identify and Reach Niche Markets for Your Business.
2. Highlight your competitive advantage. One of the keys to all advertising is to accentuate the pros of your company, those factors that give you your competitive edge. Too many ads are clever but fail to sell the benefits of the product or service.
3. Establish an image. You can recognize the McDonald's arches while whizzing by on the highway. Likewise, there are plenty of products that you recognize by their packaging or logo. Image counts when it comes to advertising and promoting your business. Too many advertisers do not work to build a consistent image. Check out Three Brand Identity Myths That Will Bring Your Business Down for additional issues to avoid.
4. You have to spend money to make money. There are ways to save money, but typically advertising is not the place to cut corners. It will affect sales, and that affects the bottom line. Successful advertising may cost some money, but that is because it works. Check out More Bang for Your Advertising Buck for cost-cutting tips that won't cut your goals.
5. Advertise in the right places. Your favorite magazine, radio station, or even television program might not be a favorite of your audience. Know what they read, watch, and listen to, and advertise in media that reaches your target market.
6. Don't allow your budget to run your advertising campaign. If you budget $5,000 per month for advertising, you've made it very easy from a bookkeeping perspective. However, if like most businesses you have seasonal highs and lows, you are spending too much money advertising during down times and not enough when you want to attract customers. Too many entrepreneurs do not budget according to their seasonal advertising needs.
7. Diversify. It is all too common for business owners to choose the best place to advertise based on price and potential rate of returns and then stop. As is the case with investing, you do not want to put all of your eggs in one basket. Spread your advertising dollars around.
8.Don't try to be everything to everyone. No product or service will appeal to everyone. Many business owners, including corporate executives, try to come up with ways to reach every market. Typically, this does not work. It can spell disaster for small businesses, who cannot afford to spread themselves too thin. Therefore, find your market and be everything you can be to that audience.
9.Test your ads in advance. If you have the time or money to invest in focus groups, you should test your ads on other people. Do they understand and accept the message that you are trying to convey? For further information, read Focus Groups: How They Can Work for Your Small Business. There are other less-expensive ways to test your ads as well: questionnaires, for example. The article Creating Questionnaires for Gathering Market Research can be helpful.
10.Monitor your ads. It is very easy to ask new customers or clients where they heard about you. As simple as this is, many entrepreneurs do not bother to do so. It is advantageous to know which ads generate business.

Increasing Web Traffic
Pick one or two things and study them in depth. You need to understand it before you can move on. So, make sure you take the time to learn the first thing you pick.
Below I list the ways that work best today and in the order you should perform them. Once you see them go to a search engine and start to study them. Remember there is much more to this than you will find on the first page of search results.
Pay Per Click
One of the best things you can do is create a Pay Per Click (PPC) account on Google or Yahoo. If you're avoiding PPC because you've been burned by it or because you're afraid of the expense, consider this:You're spending money anyway (in advertising) but PPC generally drives warm leads to your business instead of browsers. If you put a dollar value on your time, how much would you have already spent just to "tweak" the keyword percentage on your site? You have control over all kinds of factors with Google AdWords to help you control your expenses. (In fact, you should cap the amount you spend per month while you're just getting started). And, most importantly, you can tweak your advertisement periodically to see what pulls better.
Keywords
Even though you just read my scathing criticism of keyword usage in a previous paragraph, let me clarify that keywords are important to have on your site. Here's one smart way to use them.If you haven't done this lately, go and see what your competitors are doing on their site for keywords. Google your keyword and pick the top 5 sites. Check out their site's source code to discover what keywords they are using in their meta tags and headers.
Choose the top keywords and use those in your meta tags and headers. Remember that the robots that read and rank your site read from the top left to the bottom right, so put your keywords on both places. (The "keyword" currently read by robots on the bottom right of most websites is "copyright"!).
Are there other keyword strategies? Yes. And you should make sure that your keywords are used in your text. But remember that although robots rank your site, it's your human visitors who are going to give you money for your product. So make sure you're ultimately writing for them!Links
Links can be a good solution, but many small business websites use link strategies poorly. We could spend an entire article talking about link strategies (and we will) but right now, you should know this: If you are going to get someone to link to you, make sure that they have a page rank that is higher than you and that their subject matter is related to you in some way. The page rank is important for the search engines to drive you higher but the similarity that you may have with the other site will help you get customers!Blogs
Blogs are powerful tools and can help you generate links and content. Blogs are just starting to come into their own as a corporate tool and in many cases, corporate blogs are still uncharted territory. Create an offsite blog but resist the temptation to post one or two highly self-promotional posts and a link. Again, your blog might help you get higher results in a search... but it won't help you convince your customers to buy from you!i hope this article will help u a lot in increasing web traffic of your website.

Apart from being fun they make your car stand out from the rest, offering you an opportunity for self-expression and imagination in an otherwise fairly straight-laced part of life. private number plates need n?t cost the earth either. Many are available for as little as less than ?100.
Did you know that private number plates can actually rise in value? Now there's a valid reason to invest in one! Providing you select your plate carefully then you will be able to sell it at a profit in the future. The key when selecting your plate is to consider how much you are prepared to spend and then take a look at all available car registrations in this price range. You don't want anything too obscure or you will not attract any interest when it comes to selling, but if it is rare then the value can rise significantly. The sale value can be influenced by how the market is performing at the time when you want to sell and if your plate is of interest to others.
The most popular forms of private number plates are those that represent names and initials. Name number plates, especially first names, are the most desirable and therefore the most expensive of all number plates. Prices for good name number plates can be very high and relate to the popularity of the name, the match of the plate to the name and the plate style. Demand is particularly high for common first names, such as Chris, Simon, Dave and Sarah, which can sell for many thousands of pounds.

Modern Wedding Music
However, no matter what type of wedding ceremony it is, it will never be complete without the wedding music. From the wedding ceremony to the reception, wedding music is very essential. Music plays a big role in setting the mood of the celebration.
Just what exactly is a modern wedding music? About 2 years ago I was fortunate enough to have a song that I wrote for my wedding professionally recorded. Little did I know that there was a high demand for moment specific wedding songs that had a "modern" sound and feel. The standard for most weddings is to choose wedding music based on what has already been used and quite frankly, most of these songs are not "wedding" specific. My songs were written with the intention of enhancing these special moments...not highlighting popular songs from the past.
ivorytreasures is the best place if u looking for Ivory Treasures wedding music, wedding songs, download new wedding music -Ivory Treasures Wedding Music Wedding Song jusy goto www.ivorytreasures.com and find the best service.
Looking for modern wedding music doesn't have to be a difficult process if you know just where to look so just look it here.
Bathroom Remodeling Process
We are trusted bathroom remodeling service provider .
It is having said that bathroom is one of the most important parts of a home. A start for a furnished home is particularly started from main corridor and ends up in bathroom, so maximum of the homeowner emphasize more on their bathroom to have a featured look along with featured facilities like they have in their other part of the home and we make a significant effort in trying to make it true.
1.Steps to Bathroom Remodeling
Define Your Small Bath Needs and Desires
First, decide whether this will be a powder room or a full bath. Powder rooms have little more than a toilet, sink, and minor electrical work to contend with. But when you bring in the increased plumbing requirements of a bathtub or shower, you also bring in requirements for other things such as moisture protection (do you have the appropriate type of flooring?), ventilation, sizing requirements, additional bathroom cabinetry, and so on. After that, it helps to define who will be using the bathroom. If this is a full bath that will act as a master bathroom, you may need to go “all out” in terms of cabinetry and higher-end materials. After all, you will have to look at this bathroom at least twice a day for many years. But if this bathroom is for overnight guests, you can probably skimp on the quality of materials. You will most definitely not need increased storage space for guests, either.
2.Plan Your Small Bathroom
One misconception we often hear about small bathrooms: “Well, if they’re so small, I can probably map the whole thing out in my head, right?” Wrong. Small bathrooms do need extensive planning. One reason is because stock bathroom cabinets come in pre-determined sizes. You’ll probably need to work around stock cabinet sizes (or order custom cabinets). Obtain home design software that will allow you to plan your bathroom in detail.
3.Decide Whether to Hire a Pro or DIY
A small bathroom renovation, performed by a remodeling contractor and using contractor-quality materials, might cost you as little as $15,000. Doing the same remodel DIY might cost the cost by 50%. One option is to do as much as possible by yourself: demolition; basic drywall installation; flooring; and painting. Then, leave the rest to the pros. You’ll need to talk with the bath remodeling contractor to coordinate with him.
4.Strip Down to the Basics
Strip down? We mean the bathroom, not you! The degree of bathroom demolition you do depends on the scale of your project, of course; but you can expect to strip down a small bathroom in three days, probably even less. Make sure you have other bathroom facilities available, because this one will be completely disabled. Rent a roll-off dumpster for your driveway or have a hauling company take it all away. If your bathroom pre-dates 1978, be careful for lead-based paint dangers.
5.Install Plumbing and Electrical
Electrical and plumbing go in first (remember, the walls are open). Rough-in the plumbing for toilet, shower, and bathtub. Have the electrician bring in the wiring, too. You will need to have two inspections done: first, at the rough-in stage; second, after the walls have been closed up.
6.Install Flooring, Cabinets, Etc.
If you are partially DIY’ing this job, you might decide to step in at this point and take over. With plumbing and electrical fully installed, you can lay down flooring, hang cabinets, paint the walls, and install towel bars, mirrors, and that finishing touch—the toilet roll holder!
When you finally decide in your mind that you are of going for Bathroom Remodeling you have many things to look over. You have to get in touch with a reliable plumber who can fit the furnishings and fixtures. You also need to make contact with a mason if you have some tiling job. Then, there is a painter who will have to paint the total ceiling and walls, if tiles are not there. Thus in a nutshell there are many people that you require to move ahead in the job. Thus, to sum up, you have much of work to do and this will consume a lot of time and fortune of yours. But, if you opt for bathroom remodeling services all the work mentioned above will be done by the Bathroom Remodeling service providers themselves.

Silly Bandz
Silly Bandz – a great dropshipping product
Silly bandz need no introduction as they already have created a great mark on the whole world and people all around the world especially kids really like to wear them. If we name products which have become really famous in no time then silly bandz would be one of them. So, if you are looking to dropship a product which has great demand then silly bandz could be a perfect choice for you.
One of the success strategies in dropshipping is to develop a niche and silly bandz could be a really profitable niche. They are a great attraction not only for the kids but for everyone. Their different shapes, designs and colors are really famous now. People are always searching for silly bandz which no one else has. As an intelligent dropshipper what you can do is to take orders from the customers about what shape and color of silly bandz they need and then contact the manufacturer to provide those specific shapes to them. No doubt silly bandz are not a very expensive product and they are mostly available from different toy stores but its craze is so much among the people that they are ready to pay a high price to get the ones of their own choice. Therefore, dropshipping silly bandz could really be a profitable niche as there is huge opportunity to become profitable.
The craze of wearing silly bandz is not going to end so soon because a silly band could be given the shape of any famous thing. It could be given the shape of a famous cartoon character, an upcoming movie character, a super hero, or a favorite sports. So, all an intelligent dropshipper needs to do is to stay updated with the latest demand in the silly bandz market and dropship those silly bandz which people are looking for.
Gel
A gel is a solid, jelly-like material that can have properties ranging from soft and weak to hard and tough. Gels are defined as a substantially dilute cross-linked system, which exhibits no flow when in the steady-state.[1] By weight, gels are mostly liquid, yet they behave like solids due to a three-dimensional cross-linked network within the liquid. It is the crosslinks within the fluid that give a gel its structure (hardness) and contribute to stickiness (tack). In this way gels are a dispersion of molecules or particles within a liquid in which the solid is the continuous phase and the liquid is the discontinuous phase.
Gel electrophoresis tips and troubleshooting
Laemmli separating gel buffer - Buffer chloride concentration (not pH) greatly affects separation. High chloride improves stacking, but smaller proteins run with the front. Low chloride facilitates separation of small proteins at the expense of stacking. Adding HCl volumetrically from reagent con HCl is simpler and more reliable than adjusting with a pH meter.
Sample buffer - Sample (cracking) buffer has little affect on separation. pH 6.8 Tris buffer is very poor for reducing proteins (the pH is about 4.5 at 100oC!). Choose a buffer with a higher pH at 100oC. more sample tips
Casting stacking gel - Pour the stacking gel solution, then warm it with a hair dryer. This gives uniform and "complete" polymerization.
Mild cleaning solution - 1% SDS + 1% oxalic acid (overnight soak) cleans scale, rust, fingerprints and traces of grease from gel plates. Wash plates thoroughly before soaking. SDS gradually hydrolyzes; the solution becomes turbid in about six months.
Gel thickness - Minimizing gel thickness facilitates staining. With silver development, staining time increases with roughly the square of gel thickness. The thinnest available teflon sheet is 0.75mm. Thinner gels would be hard to handle. On the other hand 0.5mm or 0.5mm teflon sheet might improve silver staining substantially!
Advantages of silver staining - Silver development ("staining") is more sensitive than Coomassie Blue or ethidium bromide. It is quick and cheap (primary cost, as with Coomassie Blue, is organic solvents). Staining is "permanent"; cheap flatbed scanners record data. Commercial staining kits are superfluous. Silver detects more components giving a clearer idea of sample composition (of course one may be happier not knowing).
Protein thiols - One normally reduces proteins before SDS gel electrophoresis. Proteins with several cys residues can reoxidize during electrophoresis creating "smears". Large thiol rich proteins (e.g. serum albumin and rubisco L subunit) are especially prone to reoxidation. Reducing with DTT (see advice) and alkylating with iodoacetamide or N-ethylmaleimide prevents this.
Stability of polyacrylamide - I recently ran a Laemmli gel after it sat on the bench at room temperature for about a month. The gel appears to have partially hydrolyzed. It ran slowly (electroendosmosis). I stopped it when the dye front was ~3/4 of the way down. The gel had swollen appreciably, particularly below the "front" (higher pH region). Despite the apparent presence of carboxyl groups, the gel stained satisfactorily with silver though the surface was messy and the gel was brittle.
Polyacrylamide vs. agarose - Polyacrylamide resolves better (smaller pore size) and gives less background staining. Pore size limitations prevent polyacrylamide separations of larger macromolecules. Agarose is more convenient. Literature exaggerates it's purity.
Reproducible electrophoresis - Geometry and buffer composition determine electrical resistance of electrophoretic systems. Assuming constant geometry, at a given voltage current should be the same from gel to gel (a handy quality control parameter). Buffer variation leads to current irreproducibility. See weigh and pour recipes
Fluorescence detection - Have you seen the Dark Reader? I hoped that media such as polycarbonate centrifuge tubes and agarose would fluoresce less (reduced background fluorescence) with blue illumination - no such luck. Molecular Probes makes many useful dyes including Sypro ruby for proteins and Sybr Gold for staining nucleic acids in gels.
Gel problems
No SDS in wells - protein precipitates as it enters running gel
High salt concentration in some samples - "wide lanes" and localized distortion
Nothing on gel - failed to load or ran the wrong way (be sure to follow tracking dye as it begins to migrate).
Gel swells abnormally and is brittle - aging (polyacrylamide has partially hydrolyzed).
Gel is gooey and sticks to glass - you forgot crosslinker
Silver development slow - add more formaldehyde to developer or use fresh formaldehyde (problem may be aldol condensation, methanol is added to stabilize formaldehyde solutions, it's volatile and gradually vaporizes. Formaldehyde also oxidizes and is light sensitive.)

Signs & Symptoms of Rheumatoid Arthritis
Health is the general condition of a person in all aspects. It is also a level of functional and/or metabolic efficiency of an organism, often implicitly human.
7 Basic Everyday Health Tips
1. Move extra: Make it a daily challenge to find ways to move your body. Climb stairs if given a choice between that and escalators or elevators. Play with your kids; throw balls with friends, trim the lawn. Anything that moves your limbs is not only a fitness tool, it’s a stress buster. Think ‘move’ in small increments of time. It doesn’t have to be an hour in the gym. But that’s great when you’re up to it. Meanwhile, move more.
2. Cut Fat: Avoid such as fried foods, burgers and other fatty meals. Dairy products such as cheese, cottage cheese, milk and cream should be eaten in low fat versions. Nuts and sandwich meats, mayonnaise, margarine, butter and sauces should be eaten in limited amounts. Most are available in lower fat versions such as substitute butter, fat free cheeses and mayonnaise.
3. Give up Smoking: Smoking is harmful to your health.
4. Reduce Stress: Easier said than done, stress busters come in many forms. Some techniques recommended by experts are to think positive thoughts. Spend 30 minutes a day doing something you like. (i.e; walk on the beach or in a park; read a good book; visit a friend; play with your baby; listen to soothing music; watch a funny movie. Get a massage, a facial or a haircut. Meditate. Count to ten before losing your temper or getting aggravated. Avoid difficult people when possible.
5. Protect Yourself from Pollution: If you can’t live in a smog-free environment, at least avoid smoke-filled rooms, high traffic areas, breathing in highway fumes and exercising near busy thoroughfares. Exercise indoors in air conditioning when air quality is good. It’s a good pollution and dirt from the street deterrent.
6. Floss Your Teeth: Recent studies make a direct connection between longevity and teeth flossing. Nobody knows exactly why. Perhaps it’s because people who floss tend to be more health conscious than people who don’t? Thought for the day: Floss and be your body’s boss.
7. Keep a optimistic Mental point of view: There’s a definitive connection between living well and healthfully and having a cheerful outlook on life.
Preventive care is a valuable tool in maintaining good health. By having regular medical check-ups we can often detect problems and take care of them before they become serious enough to be life-threatening.
What causes rheumatoid arthritis?
The cause of rheumatoid arthritis is unknown. Even though infectious agents such as viruses, bacteria, and fungi have long been suspected, none has been proven as the cause. The cause of rheumatoid arthritis is a very active area of worldwide research. It is believed that the tendency to develop rheumatoid arthritis may be genetically inherited. It is also suspected that certain infections or factors in the environment might trigger the activation of the immune system in susceptible individuals. This misdirected immune system then attacks the body's own tissues. This leads to inflammation in the joints and sometimes in various organs of the body, such as the lungs or eyes.
Regardless of the exact trigger, the result is an immune system that is geared up to promote inflammation in the joints and occasionally other tissues of the body
Signs & Symptoms of Rheumatoid Arthritis
Morning Stiffness
· Morning stiffness is when a person wakes up frozen in place, feeling stiff, and has pain for at least an hour. This is one of the most common symptoms of RA, and is different from osteoarthritis, which usually only affects a person for a half an hour. When this freezing occurs, the person feels most of the pain in the hands, wrists, knees and feet. Many of the affected joint areas feel swollen and warm. Movement may become easier after stretching. Some people also have the accompanying symptoms of fever, weakness and fatigue.
Stiff Neck and Jaw
· Some people affected by RA lose movement in the neck (cervical spine) and feel stiffness to the point of restricted movement. Inflamed ligaments around the spinal cord or problems with the nervous system may cause this. Many people also experience their jaws freezing up when attempting to eat or drink.
Frozen Shoulders
· RA freezing can also affect the shoulders. This part of the body is not used as often as other areas, such as the arms and legs, and can freeze up quickly due to the inactivity. When the shoulders freeze up, all motion in the upper chest area is restricted and may cause difficulties in daily activities, such as driving or housework.
Loss of Use of Wrists and Hands
· Most people affected by RA have frozen joints in the wrists. This can affect daily activities, such as working. Fingers and knuckles often freeze up with RA and become unable to fully open or bend. Fingers may appear dislocated if the tendons slip out of place. Typically, only the joints in the middle of hand and fingers freeze, while the tips of the fingers do not.
Loss of Movement in Knees, Feet and Ankles
· Knees may freeze up with RA, restricting the movement between sitting and standing. This may occur after sitting or inactivity for a long period of time. The sole of the foot can also feel numb while walking because the joints in the middle of it have frozen up. Toes may freeze up like fingers and appear dislocated as well.
